Exploration of the Relationship of Varicella-Zoster Virus (VZV) and Alzheimer’s Disease (AD)

Introduction: Alzheimer’s disease (AD) is a neurodegenerative disorder with a multifactorial etiology. Among the proposed causes, a viral origin has been considered, particularly involving Herpes Simplex Virus type 1 (HSV-1) and Varicella-Zoster Virus (VZV). Reactivation of VZV in individuals over 65 years of age has been suggested as a potential trigger of neuroinflammation, which may contribute to the development of AD. Objective: To analyze the existing scientific literature in order to evaluate a possible relationship between VZV reactivation and the development of Alzheimer’s disease. Methods: A systematic review was conducted following the PRISMA guidelines, based on published studies related to VZV and AD. Results: VZV reactivation can lead to various alterations in the central nervous system, including dementia, through mechanisms such as neuroinflammation, vasculopathy, and direct neuronal damage. Conclusions: Although progress has been made in understanding the impact of VZV on the central nervous system, further research is needed to clarify its role in neurodegenerative diseases such as AD. Current evidence does not confirm a definitive link between VZV and Alzheimer’s disease, but neither does it exclude its possible involvement.
